离子色谱快速检测壳寡糖含量

目的:为了快速准确的进行壳寡糖样品的检测,建立一种分离度好、灵敏度高的壳寡糖离子色谱快速检测方法。方法:实验选用Dionex-CarboPac PA104×50 mm离子交换色谱柱,通过优化柱温、淋洗液配比及浓度等色谱条件,将不同聚合度的壳寡糖在30 min内实现分离。分离后的壳寡糖样品通过计算聚合度为壳2~6糖的含量,检验其样品中壳寡糖的纯度。结果:样品浓度在2.00~10.00 μg·mL-1范围内线性关系良好(r≥0.999 0),精密度实验RSD在0.1%~1.6%之间,样品加标回收率93.2%~105.0%。3种壳寡糖样品中2~6个聚合度壳寡糖的含量之和分别为25.944 μg·g-1、26.406 μg·g-1、25.374 μg·g-1,计算得到纯度为86.5%、88.0%和84.6%,符合国家标准要求。结论:该方法需要样品量少,检验快速,无需衍生,为壳寡糖质量分析、组分分离提供了依据。

Objective: For the rapid and accurate detection of chitosan oligosaccharide samples, a rapid analysis method with good resolution and high sensitivity was established.Methods: Using Dionex-CarboPac PA104×50 mm ion exchange column, different degrees of polymerization was separated in 30 min by optimizing the column temperature, the ratio and the concentration of eluent.Calculate the content of isolated chitosan oligosaccharide(DP=2-6) to examine the purity of chitosan oligosaccharide.Results: The linear relation of each component was excellent with the range of 2.00-10.00 μg·mL-1(r≥0.999 0), and the relative standard deviations of precision were between 0.1%-1.6%.The recovery rates of the method were between 93.2%-105.0%.The contents of 2-6 degree of polymerization of Chitosan Oligosaccharide in three samples were 25.944 μg·g-1, 26.406 μg·g-1 and 25.374 μg·g-1.So the purities were calculated to be 86.5%、88.0% and 84.6%, and conformed with the national standard.Conclusion: The method detects quickly using few samples and provides the reference for quality analysis and component separation of chitosan oligosaccharide.
